Basically the SMB Protocol, which stands for Server Message Block, is a protocol for sharing files and printers etc, between computers and servers on a network.

Computers connect to each server using protocols like TCP/IP and once they have achieved a connection they are then able to send commands (Server Message Blocks) to the remote server that allows them to access files etc.

SMB is also used as the foundation for Microsoft's Distributed File System (DFS) implementation.
